Objectives: The study aims to establish the standard curves for quantifying the concentrations of LncRNA IFI6lnc2 and EPB41L4A-AS1. Methods: The absolute quantitative real-time PCR method was employed to construct the standard curves based on standard samples derived from recombinant plasmids containing the coding genes for these lncRNAs. Results: The coding genes for lncRNAs IFI6lnc2 and EPB41L4A-AS1 were cloned successfully in pJET1.2 by conventional recombinant methods. The recombinant plasmids exhibited high concentrations, with pJET-IFI6lnc2 at a concentration of 1.8 x 1010 copy/μl and pJET-EPB41L4A-AS1 at a concentration of 2.4 x 1010 copy/μl. Standard curve equation of lncRNA IFI6lnc2: Y=-3.408X + 35.989 (R2=0.998, E =96.5%) and lncRNA EPB41L4A-AS1: Y= -3.4706X + 36.225 (R2=0.999, E =94.0%). Conclusion: The results of the standard curves demonstrated a linear correlation and high amplification efficiency.
