SIMULTANEOUS DETERMINATION METHOD OF 6-SHOGAOL, 6-GINGEROL, 8-GINGEROL, AND 10-GINGEROL BY HIGH-PERFORMANCE LIQUID CHROMATOGRAPHY

Abstract
Objectives: To develop a method to simultaneously quantify 6-shogaol, 6-gingerol, 8-gingerol, 10-gingerol in ginger using high-performance liquid chromatography - HPLC for application in raw material testing and finished products. Methods: The validation was carried out in accordance with ICH guidelines. Results: The optimal condition of parameters was: InertSustain AQ - C18 column (150 x 4.6 mm, 5µm); injection volume (10 µL); the mixture of ACN (C) and acetic acid 0.1% (A) as mobile phase in gradient elution mode: (0 - 10 min, C 50%; 10 - 40 min, C 50% - 90%; 40 - 45 min, C 90% - 100%; 45 - 50 min, C 100% - 50%) with a flow rate of 1 mL/minute); detection at 280nm. The method was validated with the following criteria: System suitability, selectivity, linearity, precision, accuracy, LOD, and LOQ. Conclusion: The simultaneous determination of 6-shogaol, 6-gingerol, 8-gingerol, and 10-gingerol was fully validated in accordance with ICH.
