EXTRACTION OF DIHYDROMYRICETIN AND TOTAL FLAVONOIDS FROM THE DA DAY HD HERBAL FORMULA
Main Article Content
Abstract
Objectives: To identify a suitable method for extracting dihydromyricetin (DMY) and total flavonoids (TFs) from the Da Day HD herbal formula. Methods: An experimental study was conducted to sequentially investigate the factors, including extraction method, ethanol concentration, solvent-to-herbal material ratio, extraction temperature, and extraction time. The effects of extraction method, ethanol concentration, solvent-to-herbal material ratio, extraction temperature, and extraction time were investigated. The extraction efficiencies of DMY and TFs were used as the evaluation criteria; DMY and TFs were quantified by high-performance liquid chromatography (HPLC) and ultraviolet-visible (UV-Vis) spectrophotometry, respectively. Results: With 50% ethanol, ultrasound-assisted extraction produced a higher DMY extraction efficiency than reflux extraction (80.39% vs. 74.06%), whereas the extraction efficiencies of TFs were comparable. Reflux extraction was selected because it was more amenable to scale-up. Increasing the solvent-to-herbal material ratio from 10:1 to 30:1 mL/g increased DMY extraction efficiency from 74.06% to 90.43%, whereas the extraction efficiency of TFs increased only from 92.10% to 95.51%. A ratio of 10:1 mL/g was selected to balance extraction efficiency and process feasibility. Extraction at the boiling temperature of 50% ethanol (80 - 82°C) produced higher efficiencies than extraction at 60 ± 2°C. Extending the extraction time from 2 to 3 hours only slightly increased DMY extraction efficiency but decreased TF extraction efficiency. Conclusion: The suitable conditions comprised a single reflux extraction with 50% ethanol at its boiling temperature, using a solvent-to-herbal material ratio of 10:1 mL/g for 2 hours.
