FACTORS ASSOCIATED WITH cffDNA CONCENTRATION IN IN VITRO FERTILIZATION PREGNANCIES

Abstract
Objectives: To evaluate the association between maternal age, maternal body mass index (BMI), gestational age, and cell-free fetal DNA (cffDNA) concentration in in vitro fertilization pregnancies. Methods: A retrospective cross-sectional study was conducted on 1,000 IVF pregnancies undergoing non-invasive prenatal testing (NIPT) at the Military Institute of Clinical Embryology and Histology, Vietnam Military Medical University, from January 2023 to February 2026. Results: The mean maternal age was 30.8 ± 5.1 years; the mean BMI was 21.2 ± 2.0 kg/m²; and the mean gestational age was 10.8 ± 1.2 weeks. The overall mean cffDNA concentration was 16.2 ± 5.2%. No statistically significant differences in cffDNA concentration were observed across maternal age groups (p = 0.577) or maternal BMI categories (p = 0.859). The cffDNA concentration increased with gestational age, from 14.9 ± 4.6% at 9 weeks to 17.1 ± 5.1% beyond 12 weeks (p = 0.005), with a Pearson correlation coefficient of r = 0.1701 (p < 0.001). Conclusion: In IVF pregnancies, cffDNA concentration increases significantly with advancing gestational age. Maternal age and maternal BMI are not significantly associated with changes in cffDNA concentration.
