CHARACTERISTICS OF EXOSOMES SECRETED FROM PRIMARY UMBILICAL CORD-DERIVED MESENCHYMAL STEM CELLS

Abstract
Objectives: To isolate and characterize exosomes (EXs) from umbilical cord-derived mesenchymal stem cells (UCMSCs) for application in regenerative medicine studies. Methods: 2D culture of primary UCMSCs was used to secrete EXs. EXs were then isolated by differential ultracentrifugation and characterized using transmission electron microscopy (TEM), immunoblotting (Western Blot), and pulse impedance measurement. Results: UCMSCs in 2D cultures, which were used to secrete EXs, expressed typical fibroblast-like morphology, showing positive biomarkers (> 95%) and negative markers (< 2%). The average secretion yield was 360.748 ± 59.96µg of exosomal protein/32.267 ± 2.12 × 10⁶ cells. The obtained EXs have the following characteristics: cup-shaped morphology; expression of CD63 and CD9 markers; an average size of 79 ± 19.5nm, mode value of 71nm, and a range of 55nm to 234nm. The average zeta potential of the EXs is -16mV, and the mode is -10mV. Conclusion: EXs isolated from 2D-cultured primary UCMSCs possess all the typical characteristics of EXs, suitable for further testing.
