Abstract

Objectives: To evaluate the objective response rate (ORR) and time on treatment (ToT) of patients with advanced, metastatic stage non-small cell lung cancer (NSCLC) with ALK gene mutations treated with first-line ceritinib; and to assess some side effects of the drug. Methods: A retrospective cohort study was conducted on 41 patients with advanced, metastatic stage NSCLC with ALK gene mutations treated first-line with ceritinib from 2021 to 2025 at the National Lung Hospital. Results: The average age was 52.02 ± 12.09; the male/female ratio was 1.05/1. The ORR was 90.2%, with partial response being the most common (75.6%); the median ToT was 28.48 (95%CI: 11.93 - 45.03) months. The most common side effect was diarrhoea, occurring in 41.5% of patients; elevated liver enzymes were observed in 27 patients (65.9%), mostly grade I. Conclusion:First-line ceritinib treatment in patients with advanced metastatic stage NSCLC with ALK mutations resulted in a high ORR (90.2%), a favorable median ToT (28.48 months), generally mild side effects.
